Here are the notes that I took from this intense 2 day seminar.  Most of them are standalone ideas that deserve time to absorb:

What do you expect from the seminar? If you don’t know what you expect, how will you know that you arrived there? We must define what we want.

Essence of genius is simplicity. Know the definition of words.

A professional is one who thinks it is simple.

What is truth? Exact time, place, form and event. Does not contain continuation.  A Lie is the opposite time, place, form and event, and continues.

People are ethically good. We are the worst judge of ourselves, so when we do something bad, we feel bad. When we do something good, we feel good.

Must understand the mind – everything you deal with are minds.

How do you find a purpose, how do you find what your purpose is?  It is your Fuel of the soul.  What is your purpose?  Write down your purpose.  Look up the definition of purpose.

Ideas are conveyed by sentences.  Sentences are conveyed by words or symbols. If you do not understand the sentence it is because there is a word or words that you do not know in the sentence.  If you do not understand the sentences, you will not get the idea. Your IQ is defined by the number of WRONG words you know.  Increase the number of words that you do know, and you increase your IQ.

People become criminal because of the number of words that they don’t understand. A good man comes across a word or symbol that he does not understand, which causes confusion, which causes sins. Once you go past a misunderstood word without figuring out what it means, your understanding goes away. The more words you do not understand, the stupider you become. Learn definitions of words from the dictionary.

The first dictionary was written by Webster in order to empower people to understand words so they have power and cannot be cheated.

Ethics – ability of the individual to uphold the moral code.

Justice – when an individual violates the moral code, the group puts force to get the individual to be ethical.

Moral code – a set of standards, laws, or rules that we hold ourselves to, based on our own group consensus.

If you have a problem, look at all the data of the problem in order to find a solution. For a problem to be a problem, it must contain a lie.  If something becomes complex, it contains a lie. Find the lie and you will solve the problem. Simple brings the result immediately. (Task:  change IX to 6 using just one continuous line *see solution at end)

An authority is someone who knows good P.R. Look at their result, by their actions, you can judge how they are.

Insanity is not being there – being in the past when you need to be in the present.

Spirit – this thing that knows. We need to clear false data.

Love is the flow of admiration and sympathy. Love is very close to hate, flipping very fast. Love can be used to control and cage. Use love to take the person to full potential, not cage or control.

The view point that you take defines your life. If you think you can, you can. If you think you cannot, you cannot. 99% of the people do not try, are not willing to show up.

Clever = ability to recognize truth. Smallest number in the universe is 2 = force, counterforce.

Greatness is ability to continue to love man despite all invitation to do otherwise. If you can continue to love them, no sorrow or pain. Handle the spirit, not the physical.

Prerequisite for success is to become unserious, charging toward your purpose and be sure of your success. Success is ability to experience motion, as well as the ability to control motion.

What is seriousness – when interest is intense because of penalty. Attention is stuck in the past. I better do that or else something will happen.  When you are serious, you get slow and stuck on what you do not want – one always gets what his attention is on.  You get what you validate.  Look at the problem from an unserious point of view.

Cycle of action = Start, Change, Stop.

When you start something, you also need to have closure. Exercise = make a list of ALL the open cycles that we have in our life. Do a target list with closure date, and take one or two a day. Stress is only the sum of all your open cycles.

British Royal Visit

Honoured to have been selected to escort HRH Queen Elizabeth II and HRH Prince Phillip and other dignitaries during a State Visit to Toronto.

Tent City

On behalf of our client Home Depot Canada, developed, coordinated and project managed the safe, humane and effective removal of "illegal squatters" from Toronto's infamous "Tent City", the largest and longest running illegal squat in Canadian history. Additionally, oversaw the subsequent removal of contaminated/toxic growth and the capping of 13 acres of contaminated soils from this site.

Bradley Research Group

Microarrays

The Bradley Group has worked in the micro-array area for some four years and has led to the development of a generic HT screening method for the determination of the substrate specificity of proteases, kinases and phosphatases etc. (Interfacing DNA Arrays with Combinatorial Libraries, Chem. Commun., 2005 and the last paper 2006 presents a 10,000 member peptide library). With time interest in micro-arrays has been extended to include arrays based around small molecule and polymer based transfection agents (Polyplexes and Lipoplexes for Mammalian Gene Delivery (see: Traditional to Micro-array Screening, Combinatorial Chemistry and High-Throughput Screening, 2004, 447, in which libraries of small molecules within polymers were screened for their cellular transfection abilities (delivery of DNA into cells). This is important in so called gene therapy or antisense studies. Thus there is a need to be able to efficiently deliver DNA (or RNAi) into cells. This project aims to produce new reagents, tools and methods for DNA delivery.

Stem cell control and manipulation

The Bradley group has developed so called polymer micro-arrays. This is in essence a glass microscope slide with 2,000 different polymers printed in a spot like manner across the array. This array can then be screened with mammalian cells to identify polymers that are cell selective. We are currently interested in preparing arrays that will allow cell release, of finding polymers that will allow small molecules to be placed in the array to modulate cell phenotype (physical changes). Currently, a new approach to transfect mammalian cells on polymer microarray platforms has been achieved.

Cellular Delivery

The successful and efficient introduction and delivery of materials into cells is of fundamental importance throughout many areas of biology. It is critical for the analysis of function, the perturbation of specific cellular processes, and the development of novel therapeutic strategies. The Bradley group has carried out numerous studies with small, mono-disperse, cross-linked beads (we can routinely prepare a variety of cross-linked beads from 200 nm - 5 µm) on a variety of cultured cell lines. Remarkably, and quite generally, these beads are taken up by cells These particles have a number of advantages over other approaches. Firstly, a diverse range of compounds can be attached to the microspheres including small molecule inhibitors, peptides, RNA and DNA. They are large enough to visualise using standard microscopy techniques (unlike nano-particles). They are not diluted within the cell. Populations of cells containing beads can be readily sorted automatically from other cells for subsequent analysis and it is possible to get very high uptake rates (this can be modulated through alteration of the bead size and incubation time).

HT Physical Organic Chemistry

The research group routinely uses high-thorough chemical methods to prepare chemical entities (why make one when in the same time you can make 10!). However to date little has been carried out in the area of utilising HT methods in physical organic chemistry for reaction predication, mechanistic interrogation and reaction profiling. We have begun to look at tools and chemistries to allow Hammett plots and reaction predictions to be determined by mass spectrometry with a single injection on an LCMS allowing complete interrogation and interpretation of a reaction. This research offers an approach that will enable the first steps to be taken forward for HT reaction prediction and the generation of kinetic parameters and challenges current philosophy in offering new methods orders of magnitude faster than those currently used.

Combinatorial Chemistry

Combinatorial chemistry evolved from work in the area of peptide chemistry, specially from the need to produce large munbers of products either by preparing single compounds in parallel synthesis or by preparing many compounds simultaneously in mixtures. Using combinatorial techniques these large numbers of compounds can be prepared in a faster, more efficient and cheaper way and can give rise to millions of compounds. These compounds can then be screened for the desired property (e.g. a new catalytic activity, enzyme inhibitor). Combinatorial Chemistry is now widely applied within the pharmaceutical industry as a means of identifying new leads (through random screens) and optimising the potency of feasible drug candidates (lead optimisation).
